Today I packed up all my stuff and checked out of my hotel room. I tried to get an auto to take me to “River Valley” but none of them knew where it was. I ended up walking all the way to work with all of my stuff. I really don’t have that much stuff, but carrying my backpack did make me late and sweaty when I made it to work.
We worked on optimizing the PCR and found that the Sigma column kits did not enhance the band brightness, but made the bands disappear all together. Possibly because they made me use 30 ul of H20 instead of 200ul of Elution buffer (as specified in the kit) to elute the DNA. Unfortunately I believe that Samples W6-W10 no longer contain any DNA. So they will have to be dropped from the study.
The good news was that the bands did get brighter when double the amount of DNA was added to the PCR mix. I can now proceed and do PCR for all the samples. The project is going along swimmingly : ).
I left work early to head to the train station. The train was late to the station by about an hour so I sat around at the station for quite some time. There were many pilgrims coming to a temple in Kottayam. They have coming for the past few weeks now. They wear all black, walk barefoot, and carry a parcel wrapped in cloth on their head. Apparently when they finally arrive at the temple ( I can’t remember the name ) they break a clay pot, which they carry in the cloth on their head. Anyway, it is pretty interesting to watch all the pilgrims unload in groups of 200+ people from the trains at Kottayam station.
